Your best line of defense is to use a "strong" master password, that is, one that's reasonable long, is not a word or a variation of a word that's in any dictionary, and is not something that can be guessed or found out about you (SSN, phone number, etc.). Having said that, this won't help you if you use a trivial and/or short master password, as these are this first that any attacker will try. PasswordSafe's database format is designed to be resistant to brute-force attacks, meaning that each password check takes a significant amount of computational resources.
